Antioxidant and Anti-wrinkle Effects of Quercetin 3-Glucoside-7-Rhamnoside Isolated from Hippophae rhamnoides Fruits |

ABSTRACT |
Although many studies have been performed to search materials that regulate skin aging, little is known about
the effect of quercetin 3-O-glucoside-7-O-rhamnoside (Q3G7R) from Hippophae rhamnoides (H. rhamnoides )
L. fruits. In this study, we identified Q3G7R from H. rhamnoides L. fruits, and then measured the antioxidant
activity of Q3G7R by 1,1-diphenyl-2-picryl hydrazyl (DPPH) method and its cytotoxicity in human dermal
fibroblasts using sulforhodamine B (SRB) assay. Also, we investigated effect of Q3G7R on procollagen
synthesis and TNF-α induced Matrix Metalloproteinase-1 (MMP-1) expression in human dermal fibroblasts.
As the results, Q3G7R didn’t exhibit cytotoxicity at 100 μg/mL showing cell viability over 100 % and antioxidant
activity with 56.4% at concentration of 1 mg/mL. Procollagen synthesis in human dermal fibrolasts increased
about 2 times higher than control by treatment with 1 μg/mL of Q3G7R. TNF-α induced MMP-1 expression was
reduced to 85.58 ng/mL by treatment with 1 μg/mL of Q3G7R. Therefore, these results suggest that Q3G7R
can be effective antioxidant and antiwrinkle ingredient for cosmetics applications. |
