Effect of Rosemary Extract on Antioxidative Activity and Melanogenesis in Cultured SK-MEL-3 Cells
ABSTRACT
To evaluate the effect of Rosemary(Rosemarinus officinalis) extract on antioxidative activity and
melanogenesis on cultured SK-MEL-3, the cell viability, DPPH-radical scavenging activity, superoxide
dismutase(SOD)-like activity, tyrosinase activity and a total amount of melanin were
measured. In this study, SK-MEL-3 cells were treated with the media containing 1∼20μM, hydrogen
peroxide(H2O2) for 4 hours. H2O2 significantly decreased cell viability compared with control and
XTT50 value was determined at 20μM of H2O2. In the antioxidative effect of Rosemary extract, it
showed DPPH-radical scavenging activity and SOD-like activity. While, in the melanogenic effect of
Rosemary extract, it significantly decreased the tyrosinase activity and a total amount of melanin
which were increased by H2O2. From above the results, it is suggested that Rosemary extract was
effective in the protection of H2O2-induced cytotoxicity by antioxidative activity and inhibition of
melanogenesis.
