Effects of Alliin on Cellular Protection, Up-regulation of Collagen and Down-regulation of MMP1 in Human Dermal Fibroblasts

ABSTRACT
The attention to aging has been reflected in various aspects. In the cosmetics industry, the development of anti-aging products has been in a steady progress. Anti-inflammatory and antioxidant research has been conducted using garlic extract, but relevant research and development have been insufficient due to the difficulties to use garlic as a cosmetic ingredient due to its characteristics such as the distinctive scent of garlic. Thus, based on the need for development of an ingredient to complement the weaknesses, alliin was selected as the research subject as it is heat-stable and also its activity is maintained in the dry state. The present study aims to examine the effect of alliin in inhibition of aging by giving normal human dermal fibroblast (NHDF) H2O2-oxidative stress. In order to test the efficacy of alliin, 3-(4,5-dimethylthiazol-2-yl)-2,5-dipheyltetrazolium bromide thiazolyl blue assay, enzyme-linkes immunosorbent assay, and quantitative real-time polymerase chain reaction were performed. It was demonstrated that treating H2O2-treated NHDF with alliin was effective in restoring damaged cells as the treatment increase the survival rate of the cell. It was also demonstrated that alliin treatment given to the NHDF treated with H2O2-oxidative stress facilitated the generation of procollagen type I and inhibited the generation of MMP1 protein. In addition, the alliin treatment was found to facilitate the gene expression of the COL1A1 and inhibit the gene expression of MMP1. Taken together, the effect of alliin in improving aged skin was demonstrated in the cell efficacy test, and alliin is expected to have a value as an excellent anti-aging ingredient for functional cosmetics.
